Answer

Plasma half-life (t½) is the time taken for the plasma concentration of a drug (or the amount of drug in the body) to fall to half its original value.

For first-order kinetics

  • t½ = 0.693/Ke (Ke = elimination rate constant)
  • Constant, independent of drug concentration/dose
  • After about 4–5 half-lives, the drug is considered essentially eliminated (~94–97%) and steady-state on repeated dosing is achieved

Clinical significance

  • Determines dosing interval — drugs with short t½ need frequent dosing (or sustained-release formulations)
  • Determines time to reach steady state with repeated dosing (~4–5 half-lives)
  • Guides how long to wait after stopping a drug for it to be eliminated
  • In zero-order kinetics, half-life is not constant — it increases with dose (e.g., phenytoin, ethanol)
