Answer

Ondansetron is a selective 5-HT3 receptor antagonist.

Mechanism of action

  • Blocks 5-HT3 receptors peripherally (vagal afferents in the gut) and centrally (chemoreceptor trigger zone/nucleus tractus solitarius), preventing the vomiting reflex

Uses

  • Chemotherapy/radiotherapy-induced nausea and vomiting
  • Postoperative nausea and vomiting
  • Hyperemesis gravidarum (with caution)

Adverse effects

  • Headache
  • Constipation
  • QT prolongation (with high doses)
  • Flushing
