Question

Mechanism of action and therapeutic indications of Atorvastatin

Q114 marksShort Answers

Answer

Atorvastatin is an HMG-CoA reductase inhibitor (“statin”).

MOA: competitively inhibits HMG-CoA reductase, the rate-limiting enzyme in hepatic cholesterol synthesis, reducing hepatic cholesterol production; the compensatory upregulation of hepatic LDL receptors that follows increases clearance of LDL from plasma.

Therapeutic indications: hypercholesterolemia/dyslipidemia, primary and secondary prevention of atherosclerotic cardiovascular disease, acute coronary syndrome (started early, independent of baseline lipid levels — plaque-stabilizing effect).
