Malignant hyperthermia (triggered by volatile anesthetics and succinylcholine in genetically susceptible individuals) results from uncontrolled release of Ca²⁺ from the sarcoplasmic reticulum via the ryanodine receptor, causing sustained muscle contraction, hypermetabolism, and severe hyperthermia. Dantrolene sodium directly blocks the ryanodine receptor, preventing further Ca²⁺ release from the sarcoplasmic reticulum, which halts the sustained contraction and hypermetabolic state — making it the specific antidote for this condition.
