Therapeutic uses

  • Tuberculosis (first-line drug, part of combination therapy)
  • Leprosy (multibacillary and paucibacillary, part of MDT)
  • Prophylaxis for close contacts of meningococcal meningitis and H. influenzae type B
  • Carrier state elimination for meningococcus

Adverse effects

  • Hepatotoxicity
  • Orange-red discolouration of body secretions (urine, sweat, tears) — harmless but should be explained to patient
  • Flu-like syndrome (with intermittent/interrupted dosing) — fever, chills, myalgia
  • GI upset
  • Potent hepatic microsomal enzyme induction — reduces efficacy of oral contraceptives, warfarin, corticosteroids, and other co-administered drugs
  • Thrombocytopenia, haemolytic anaemia (rare, immunological)
  • Cutaneous reactions, pruritus
