Laboratory diagnosis of malaria (Plasmodium species):

  • Peripheral blood smear microscopy — gold standard; thick smear for screening, thin smear for species identification/parasitaemia quantification (Giemsa stain).
  • Rapid diagnostic tests (RDTs) — HRP-2 (P. falciparum-specific) or pLDH/aldolase (pan-species) antigen detection.
  • QBC method — fluorescence-based concentration technique, more sensitive but does not allow precise species identification.
  • PCR — highly sensitive/specific, detects low-level/mixed infections.
  • Serology — detects past exposure, not useful for acute diagnosis.
