Principle: growing the fungus on a small block of culture medium placed directly on a slide (rather than in a Petri dish), with a coverslip resting on top, allows the fungus to grow and produce its characteristic fruiting structures in situ, preserving their natural, undisturbed three-dimensional arrangement for microscopic examination.
Procedure: a small square block of an appropriate agar medium (e.g., Sabouraud dextrose agar or a specialized medium like potato dextrose agar or cornmeal agar) is placed on a sterile glass slide; the block is inoculated on its sides with the fungal isolate; a sterile coverslip is placed on top of the block; the entire slide assembly is incubated in a humidified chamber (e.g., a Petri dish containing moist filter paper, to prevent the agar block from drying out) for several days, allowing fungal growth to occur both on the agar block and up onto the underside of the coverslip.
Examination: after adequate growth, the coverslip (with fungal growth adhering to it, showing intact fruiting structures) is carefully lifted off, mounted onto a fresh slide with a mounting medium (e.g., lactophenol cotton blue), and examined microscopically — the fungal structures are seen in their natural, undisturbed spatial relationship, which is critical for correctly identifying characteristic diagnostic features such as conidiophore branching patterns and the arrangement of macroconidia/microconidia.
Clinical significance: particularly valuable for identifying dermatophytes and other moulds where precise conidial arrangement (rather than just conidial shape alone) is a key diagnostic feature, providing more reliable species-level identification than a disturbed/teased-mount preparation from a standard plate culture.
