Answer

Griseofulvin is an antifungal active against dermatophytes.

Mechanism of action

  • Deposits in keratin precursor cells of skin/hair/nails
  • Binds to fungal microtubular protein, disrupting mitotic spindle formation and inhibiting fungal cell division (fungistatic)

Uses

  • Dermatophytosis (tinea infections of skin, hair, nails) — not effective against Candida or other yeasts

Adverse effects

  • GI upset, headache
  • Hepatotoxicity
  • Photosensitivity
  • Disulfiram-like reaction with alcohol
  • Enzyme induction — reduces efficacy of oral contraceptives, warfarin
  • Contraindicated in pregnancy (teratogenic) and porphyria
