Negative staining is a microscopy technique in which the background is stained while the microorganism/structure itself remains unstained (colourless), appearing as a clear/bright object against a dark background — the reverse of conventional positive staining, where the organism itself takes up the dye.
Principle: an acidic dye (e.g., India ink, nigrosin, or Congo red) carries a negative charge; since the bacterial cell surface is also typically negatively charged, the dye is repelled from and does not penetrate or adhere to the organism, instead depositing in the surrounding background/medium — the organism stands out as a clear, unstained area against the opaque, dark-stained background.
Applications:
Demonstration of bacterial capsules — capsules do not stain well with ordinary basic dyes and are also often washed away or distorted by heat-fixation used in conventional staining; the India ink negative-staining technique demonstrates the capsule as a clear halo surrounding the (separately counterstained or unstained) cell body against the dark background, without the distortion caused by heat-fixing — classically used to demonstrate the capsule of Cryptococcus neoformans in CSF, and applicable to bacterial capsules as well.
General morphological examination — allows visualization of overall cell size and shape without the distortion/shrinkage that can occur with heat-fixation and positive staining methods, useful for organisms difficult to stain by conventional methods (e.g., spirochetes, some fastidious/delicate organisms).
Advantage: since no heat-fixation or chemical fixation is required, the technique preserves natural cell size/shape and is technically simple and rapid, making it useful as a quick screening method.
