L-D bodies (Leishman-Donovan bodies) are the intracellular amastigote form of Leishmania species (e.g., L. donovani, the causative agent of visceral leishmaniasis/kala-azar), found within the cytoplasm of infected host macrophages — the diagnostic tissue form of the parasite in the mammalian host.

Morphology: small (2–4 μm), round-to-oval, non-flagellated intracellular bodies, each containing a nucleus and a rod-shaped kinetoplast (a specialized mitochondrial DNA-containing structure) lying at right angles or parallel to it — the presence of both nucleus and kinetoplast together within the same small round body is the key identifying feature on Giemsa-stained smears, distinguishing LD bodies from other intracellular structures/artifacts.

Clinical/diagnostic significance: demonstration of LD bodies within macrophages on a Giemsa-stained smear of splenic aspirate (highest sensitivity, though with a bleeding risk given splenic fragility) or bone marrow aspirate (the safer, standard alternative) is the classical, direct parasitological method for confirming visceral leishmaniasis (kala-azar) — LD bodies are typically seen packed within macrophages, sometimes with several amastigotes per cell, reflecting the parasite’s intracellular multiplication.

Naming: the term honours William Leishman and Charles Donovan, who independently first described this intracellular parasitic form (in 1903) in splenic tissue from patients with visceral leishmaniasis, leading to naming of the causative species Leishmania donovani.
